Requirement of PilJ periplasmic domain for regulation of twitching motility and cAMP in P. aeruginosa.

1. Created a truncated form of pilJ, which lacks the periplasmic domain. 2. Used homolgous recombination to replace the chromosomal copy of full length pilJ with the truncated version. After creating this strain (PilJperi) 3. Determine this mutation results in reduced twitching motility. 4. Creating a deletion of the entire pilJ gene, and test this strain for twitching motility and cAMP levels.

Tasks and responsibilities:

Note-taking, creation of strains and maintenance of the strain collection, as well as making own buffers & media. Students will also attend, and present data at weekly laboratory meetings.
